Single cell and spatial genomics
The Kennedy Institute has a state-of-the-art shared research facility to support our single-cell and spatial genomics research.
SINGLE CELL GENOMICS
The facility provides best practise end-to-end workflows to assist with cell isolation, capture and library preparation. A cassette-based Sony SH800S FACS and LUNA-FX automated fluorescent cell counter are used for identification and isolation of live cell populations. For cell capture the facility is equipped with both the 10x Chromium and BD Rhapsody platforms. Library quality is assessed with an Agilent TapeStation prior to submission for sequencing at external services such as the Oxford Genomics Centre. The facility is in routine use by Kennedy groups for multimodal single-cell profiling of gene expression, surface-protein abundance (CITE-seq) and T-cell and B-cell V(D)J sequencing in tissues of interest.

The facility is currently building up a suite of complementary technologies for spatial genomics. Our nanoString GeoMx DSP platform allows researchers to use immunofluorescence microscopy to target regions of interest within tissues for targeted or transcriptome-wide profiling. Later in 2022 the facility will bring onstream the nanoString CosMx platform, which enables analysis of up to a thousand genes with effective segmentation at the single cell-level. The facility also supports use of the established 10x Visium spatial transcriptomics workflow, in which polyA mRNAs are captured on a spatially indexed array.

The Single Cell and Spatial Facility is managed by Moustafa Attar who helps our research groups to plan and conduct their experiments. The facility is directed by Associate Professor Sansom within the Institute’s Data Science Platform.
